Studies on the Pharmacological Action of the Water Extract of "Zaezo"


Abstract
"Zaezo", a chinese drug, is a dried larva of the cicadae (Platypleura kaempferi FABRICIUS) and is widely used in Oriental Medicine as a diuretic agent. Clinical evidence has accumulated to show its effectiveness, but no attempt has been made to evaluate the nature and properties of this drug. The present report deals with certain aspects of general pharmacology of the water extract of "Zaezo."
1. Single doses subcutaneously of 0. 40.5mg. of the water extract per gm. of body weight, produces uneasiness, followed by convulsion and unconsciousness in the mouse, and the minimal lethal dose for the mouse is 0.71.0 mg. per gm. of body weitht.
2. No variation is found in the blood pressure of rabbits with a dose of 12 cc (1:100) of the extract per kg. of body weight.
3. In the isolated toad¢¥s heart, the extract (1:100 01:5000) shows a positive isotropic and chronotropic effects. The cardiotonic component of the extract is thermostable and is not destroyed in the heart.
4. The extract causes contraction of rabbit¢¥s coronary artery in 1:100, 0001:1, 000, 000; in rabbit¢¥s ear vessel in 1:10, 000: toad¢¥s lung and mesertereric vessels in 11, 000 and, on the contrary, causes slight dilatation of the vessels of the toad¢¥s hind limbs in 1:1001:1, 000.
5. In the isolated intestine segment of rabbits, the extract (1:100) reduces peristaltic movement and tonus.
6. The extract (1:1, 000) has no effects on the isolated intestine segment of toads, but produces a slightly increased tonus only in the higher concentration (1:500).
7. In the isolated uterus segment of rabbits, tonus is augmented markedly by the extract (1:1, 000).
8. The extract (1:1, 0001:10, 000) augments automatic movement and tonus of the toad¢¥s bladder.
9. A marked diuresis in accompanied with dilatation of the blood vessels of the kidney in the toad is observed by perfusing the kidney with the extract according to Bainbridge¢¥s method, but no effects we-re noted when the urinary tubules of the toad are perfused alone.
10. The remarkable diuresis is found by intravenous injection of the extract (1:1001:11,000) m doses of 12 cc per kg. of body weight both in normal and experimentally produced sublimate nephrotic rabbits.
11. The albumin reaction as well as the microscopic findings of urine in the experimentally produced sublimate-nephrotic rabbit gradually clears up and becomes normal on the successive administration of the extract (1:1001:1, 000) in a dose of 12cc per kg. of body weight.
